Analysis of the newly discovered control of HSV-1 replication by the PP2A B-type subunit PR130

Zusammenfassung:

Die Phosphatase-2A (PP2A) B-Untereinheit PR130/PPP2R3A kontrolliert die Aktivierung von Checkpoint-Kinasen und steuert somit den Ablauf des Zellzyklus und die genomische Integrität der Zelle. Virusinfektionen induzieren DNA-Replikationsstress, DNA-Schäden und verursachen die Aktivierung dieser Kinasen. Da bislang unbekannt ist, ob der PP2A-PR130 Komplex virale Replikationsabläufe beeinflusst, wurde in enger Zusammenarbeit der Projektleiter dieses Forschungsvorhaben erfolgreich etabliert.

Bisherige Daten zeigen, dass PR130 die Replikation von Herpes-simplex-Virus Typ 1 (HSV-1) im Zellkern unterdrückt und die Phosphorylierung und Stabilität von Checkpoint-Kinasen in infizierten Zellen reguliert. Basierend auf dem CRISPR-Cas9 Verfahren wird nunmehr untersucht, inwiefern PR130 die HSV-1-verursachten Veränderungen des Zellzyklus und der DNA-Reparaturmechanismen moduliert. In diesem Kontext werden zelluläre und virale Proteine mittels Proteomik-Analysen bestimmt, die durch den PP2A-PR130-Komplex kontrolliert werden. Der Einfluss klinisch anwendbarer Checkpoint-Kinase-Inhibitoren auf die Virusreplikation wird in relevanten Zellkultursystemen analysiert. Darüber hinaus wird auch der Einfluss der PP2A B-Untereinheit B56β auf die Replikation von HSV-1 untersucht.

Da bislang keine Publikationen zum Einfluss von PR130 auf die Replikation viraler Infektionserreger existieren, werden die Ergebnisse der geplanten Experimente die bestehende Datenlage signifikant erweitern.

Charakterisierung des antiviralen und möglichen entzündungshemmenden Wirkmechanismus von ProcCluster® während Influenza Virus bzw. SARS-CoV-2 Infektion

Zusammenfassung:

Zirkulierende Viren, wie immer wieder neu auftretende Influenza A Viren oder Coronaviren, hierzu gehört das schwere akute Respiratorische Syndrom Corona Virus-2 (SARS-CoV-2), stellen nach wie vor eine globale Belastung für die öffentliche Gesundheit dar. Im Allgemeinen teilen Influenza A Viren und Coronaviren das Pandemiepotenzial. Die Entwicklung effizienter Impfstoffe ist zeitaufwendig und spezifische antivirale Behandlungsstrategien sind rar. Die Zulassung von Substanzen, die bereits klinisch für andere Krankheiten verwendet werden, sind kurzfristig eine vielversprechende Option zur Behandlung von Infektionen. Hierbei sind pharmazeutische Substanzen die Virus-unterstützende zelluläre Faktoren oder überschießende Immunreaktionen hemmen bzw. Entzündungsprozesse auflösen von besonderem Interesse. Erste Untersuchungen deuten auf eine hemmende Wirkung von ProcCluster®, einem Lokalanästhetikum mit entzündungshemmenden Eigenschaften, sowohl auf Influenza A Virus als auch auf SARS-CoV-2 Vermehrung und Verbreitung in vitro hin.

Während der Förderung durch den EUROPÄISCHEN FONDS FÜR REGIONALE ENTWICKLUNG – EUROPA FÜR THÜRINGEN werden in Kooperation mit der inflamed pharma GmbH Jena die Regulation der antiviralen und möglichen entzündungshemmenden Wirkung von ProcCluster® und ProcCluster®-haltigen Produkten während Influenza A Virus bzw. SARS-CoV-2 Infektion in vitro näher charakterisiert. Die Aufklärung der molekularen und regulatorischen Mechanismen von ProcCluster® und ProcCluster®-haltigen Produkten während viraler Infektion ist notwendig, um zunächst Grundlagen zur Einschätzung des Potentials von ProcCluster® und ProcCluster®-haltigen Produkten für eine spezifische Anwendbarkeit bei Influenza A Virus oder SARS-CoV-2 Infektion zu erhalten.

COMDRUG-CoV-2: Die Wirkung häufig eingenommener Medikamente auf das alveolare Gleichgewicht während einer SARS-CoV-2-Infektion

Zusammenfassung:

Ziel des Projektes ist es, unter Verwendung eines komplexen humanen Alveolus-on-a-Chip Modells, einem Modell, das Bedingungen der menschlichen Lunge nachahmt, die Pathophysiologie der SARS-CoV-2 Infektion zu untersuchen. Dabei soll die Wirkung von Substanzen gegen allgemeine Erkältungskrankheiten, Herz-Kreislauf-Erkrankungen und überschießende Immunantwort getestet werden. Insbesondere deren Wirkung auf Vermehrung und Verbreitung von SARS-CoV-2 im Epithel und Endothel, die Aktivierung von Entzündungsparametern und Schädigung der Zellbarrieren wird analysiert. Darüber hinaus werden SARS-CoV-2 induzierte Effekte auf Endothelzellen und die von diesen ausgehende Funktionen auf das vaskuläre System näher betrachtet, da dieses bei schweren COVID-19 Verläufen zu Blutgerinnungsstörungen und Thrombenbildung führen können.

Infektiosität der Covid-19-Patienten über die Dauer der Erkrankung

Zusammenfassung:

Die Diagnostik einer Infektion mit SARS-CoV2 erfolgt über den Nachweis von genetischem Material des Virus mittels Reverse-Transkriptase-quantitativer Polymerase-Kettenreaktion (RT-qPCR). In Proben von COVID-19-Patienten lässt sich genetisches Material allerdings oft 14 Tage und länger nachweisen, obwohl die Patienten keine Symptome mehr zeigen. Die Infektiosität von COVID-19-Patienten kann über die Anzucht und Bestimmung infektiöser Viruspartikel in Zellkultursystemen mittels sog. Plaquetitration ermittelt werden. Diese Methode ist jedoch sehr zeitaufwendig und deshalb nicht für die Routinediagnostik geeignet. In dem Projekt werden beide Methoden eingesetzt, um über die Dauer der Erkrankung tatsächlich vorliegendes infektiöses Material zu ermitteln und mit den Werten der RT-qPCR zu vergleichen. Weiterhin ist es Ziel des Projektes eine Methode zu entwickeln, die den Nachweis von infektiösem Material auch in der Routinediagnostik erlaubt.

Zusammenfassung:

Es wird untersucht, ob in Verstorbenen infektiöse SARS-CoV2 nachweisbar sind und ob von diesen eine Gefährdung ausgeht. Da derzeit wenige Informationen diesbezüglich vorliegen, ist geplant, verschiedene Organe auf die potentielle Infektiosität zu untersuchen.
